Mossel et al. described improved recovery of yeasts and molds compared to acidified agar. This medium is supplemented with oxytetracycline as a selective agent, rather than relying on the low pH of acidified agar to suppress bacterial growth. Acidified agar is commonly used for detection of yeasts and molds in foods and dairy products, and OGYE Agar Base provides an alternate medium. This formula was prepared with a neutral pH, deviating from the usual acidified medium. Several investigations have found a neutral pH with the addition of a selective agent have been successful in supporting fungal growth and restricting bacterial growth. Malt Extract Agar is used for the cultivation of fungi and is not intended for use in the diagnosis of disease or other conditions in humans. An acidic medium which will support the growth of most yeasts and molds whilst inhibiting most bacteria. It was first described by Thom and Church in 1926 in a study of Aspergillus spp. claiming the high carbohydrate content ensured rapid growth. Selectivity can be increased by further lowering the pH with the addition, after sterilization, of Lactic Acid (X037). It should be noted that excess heating of this medium together with its low pH can easily result in hydrolysis of the agar gel producing soft plates. He found that an agar medium containing orange serum (juice) was superior to Lindegren Agar in isolating the microorganisms responsible for spoilage causing a buttermilk off-odor. Murdock, Folinazzo, and Troy found Orange Serum Agar, pH 5.4 to be a suitable medium for growing Leuconostoc, Lactobacillus, and yeasts. LB Agar (Miller), a nutritionally rich medium designed for growth of pure cultures of recombinant strains, is based on the formulation described by Miller. E. coli is grown to late log phase in LB Medium. Some plasmid vectors replicate to a high copy number and do not require selective amplification. Some vectors do not replicate so freely and need to be selectively amplified. Chloramphenicol may be added to inhibit host synthesis and prevent replication of the bacterial chromosome.LB Agar, Miller contains 10 g/L of sodium chloride, different from the levels in Lennox formulations.
